Item Immobilization and Characterization of Cellulase on Sepabeads and Relizyme(Видавництво Львівської політехніки, 2013) Bilgin, Ramazan; Serkan Yalcin, M.In this study, the covalent immobilizations of cellulase from Trichoderma reesei onto Sepabeads and ReliZyme were studied. The optimal immobilization conditions of the cellulase onto the mentioned supports were investigated using response surface methodology. The optimal working conditions and kinetic parameters of free and immobilized cellulase preparations were also determined. The optimal immobilization pH, immobilization time and cellulase concentration onto Sepabeads were determined as 6.5, 6 h and 1.8 mg/mL, respectively. The corresponding immobilization values onto ReliZyme were 6.5, 2 h and 1.8 mg/mL.Item Partial purıfication of catalase from the cucumber (cucimus sativus)(Видавництво Львівської політехніки, 2011) Bilgin, Ramazan; Seriner, Ramazan; Serkan, Yalcin M.In this study, catalase (E.C.1.11.1.6) activities were determined in the extract of cucumber (Cucimus Sativus) activities were determined after homogenization, ultracentrifugation and DEAEcellulose ion exchange column chromotography steps After the ultracentrifugation step, catalase activities were found as 46.51 U/mg prot. and 3.19 U/mg prot., respectively. Afterdesalting with column chromotography, catalase spesific activities were found as 393 U/mg prot. and 0.479 U/mg prot., respectively. Enzyme preparations were stored at after 96 hours catalase lost almost all of its activity.
